I have used the Quickdraw plugin over at aescripts, it's great. Do your drawing and refine strokes, with easy retiming on the fly

(I can do the same thing, probably using a similar workflow.
1: Convert art to vectors. Open in Illustrator.
2. Copy paths from Illustrator to AE. Use generated vector masks as position path for hand with pen.
3. Paste mask shape into hand layer position attributes , AE auto converts that to keyframes.
Hand will follow path shape. Composite Hand layer over artwork, use stroke effect to reveal artwork as hand passes over it.
That's the gist of it.
