I’ve seen that problem with high rez comps but a restart of AE has always resolved it. My speculation from observation is usually memory starvation issues related to the 2GB frame buffer limitation.

No audio at all. Tried multiple RAM and disk purges, reopening proj, etc...

just a check... are you ram previewing with audio?  maybe try turning off the audio on the preview panel?  I ran into something once where the audio file caused the ram preview to stop very short... 

 

I've also seen oddities like only able to preview like 10 frames of a super simple 1080p comp with no explanation..  flushing all memory and stuff, and removing audio seemed to fix it if I recall..
